Investigating specialized plant metabolites, traditionally referred to as ‘secondary metabolites’ present in leaf extracts of cashew trees (Anacardium occidentale) resistant and susceptible to anthracnose disease was carried out using metabolomics combined with chemometric tools. We used clones of dwarf-cashew with the following variations of characteristics: resistant and healthy (CCP 76, BRS 226, BRS 189), susceptible and healthy (BRS 265), and another clone also susceptible but affected by the disease (BRS 265). The UPLC-QTOF-MSE (ultra performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry) analysis of the leaves of Anacardium clones allowed us to annotate a total of 39 metabolites. The multiple orthogonal partial discriminant analysis of the least-squares (OPLS-DA) allowed twelve metabolites to be potential biomarkers of differentiation among the clones studied. Namely, the triene-(17:3)-anacardic acid found on CCP 76 and BRS 189 clones, respectively, is the main candidate for biomarker of resistance. While catechin, B-type procyanidin isomers, and procyanidin dimer monogallate identified in BRS 265 are significant potential biomarkers of susceptibility.

Resumo O inseto sugador, Glycaspis brimblecombei Moore (Hemiptera: Aphalaridae), é de origem australiana e reduz a produtividade de cultivos do gênero Eucalyptus. O parasitoide Psyllaephagus bliteus Riek (Hymenoptera: Encyrtidae) é o principal agente utilizado no manejo integrado de G. brimblecombei. Endossimbiontes, em insetos, são importantes na adaptação e proteção de seus hospedeiros ao ambiente que habitam. Wolbachia, um simbionte intracelular, induz alterações reprodutivas, como feminização, incompatibilidade citoplasmática, morte de machos e partenogênese. O objetivo foi relatar o primeiro registro de Wolbachia pipientis em populações de G. brimblecombei e de seu parasitoide P. bliteus em campo no Brasil. Ramos com adultos de G. brimblecombei e P. bliteus foram coletados em árvores de eucalipto em plantios comerciais em seis estados do Brasil e, após a emergência, os insetos obtidos foram congelados -20 °C. A reação em cadeia da polimerase (PCR) foi realizada para detectar o endossimbionte Wolbachia. Wolbachia pipientis foi identificado em indivíduos de G. brimblecombei e de seu parasitoide P. bliteus de populações de Agudos e Mogi-Guaçu (São Paulo), Itamarandiba (Minas Gerais) e São Jerônimo da Serra (Paraná), Brasil.
Abstract The sucking insect, Glycaspis brimblecombei Moore (Hemiptera: Aphalaridae), is originally from Australia and reduces the productivity of Eucalyptus crops. The parasitoid Psyllaephagus bliteus Riek (Hymenoptera: Encyrtidae) is the main agent used in the integrated management of G. brimblecombei. Endosymbionts, in insects, are important in the adaptation and protection of their hosts to the environment. The intracellular symbionts Wolbachia, induces reproductive changes such as cytoplasmic incompatibility, feminization, male death and parthenogenesis. The objective of this study was to report the first record of Wolbachia pipientis in populations of G. brimblecombei and of its parasitoid P. bliteus in the field in Brazil. Branches with adults of G. brimblecombei and P. bliteus were collected from eucalyptus trees in commercial farms in six Brazilian states and, after emergence, the insects obtained were frozen at -20 °C. Polymerase chain reaction (PCR) was performed to detect the Wolbachia endosymbiont. Wolbachia pipientis was identified in individuals of G. brimblecombei and its parasitoid P. bliteus from populations of the counties of Agudos and Mogi-Guaçu (São Paulo State), Itamarandiba (Minas Gerais State) and São Jerônimo da Serra (Paraná State) in Brazil.

Abstract BINGO (BAO from Integrated Neutral Gas Observations) is a unique radio telescope designed to map the intensity of neutral hydrogen distribution at cosmological distances, making the first detection of Baryon Acoustic Oscillations (BAO) in the frequency band 980 MHz - 1260 MHz, corresponding to a redshift range 0.127 < z < 0.449. BAO is one of the most powerful probes of cosmological parameters and BINGO was designed to detect the BAO signal to a level that makes it possible to put new constraints on the equation of state of dark energy. The telescope will be built in Paraíba, Brazil and consists of two \thicksim 40m mirrors, a feedhorn array of 50 horns, and no moving parts, working as a drift-scan instrument. It will cover a 15 ^{\circ} ∘ declination strip centered at \sim \delta ∼ δ =-15 ^{\circ} ∘, mapping \sim ∼ 5400 square degrees in the sky. The BINGO consortium is led by University of São Paulo with co-leadership at National Institute for Space Research and Campina Grande Federal University (Brazil). Telescope subsystems have already been fabricated and tested, and the dish and structure fabrication are expected to start in late 2020, as well as the road and terrain preparation.

The essential oil of Cymbopogon nardus, citronella, has been extensively studied. However, the chemical and biological properties of the ethanolic extract (EE) of C. nardus have not been evaluated. The aim of this study was to characterize the chemical composition of the EE of C. nardus and its active fraction (FrD). Moreover, the cytotoxic and antifungal properties of these extracts against Candida species with different resistance profiles to conventional drugs were evaluated. The compounds identified in EE were mono-C- and di-C-glycosyl flavones and phenylpropanoid glycosides. Phenylpropanoid glycosides were identified in FrD. EE showed antifungal activity, with minimum inhibitory concentration (MIC) values ranging from 62. 5 to 500 µg mL-1. FrD was more effective against C. glabrata, as evidenced by the lowest MIC value (15. 6 µg mL-1). EE inhibited yeast growth similar to amphotericin-B, as demonstrated by similar time-kill curves. EE inhibited C. albicans hyphae formation and mature biofilm of C. albicans, C. krusei and C. parapsilosis. The results of the chemical and biological analyses of EE and its fractions provided novel information and may contribute to control of infections caused by Candida species.

Many instrumental techniques are used to monitor the quality of food products, highlighting near infrared spectroscopy (NIRS). In order to determine the authenticity of 60 commercial oil samples seized by the Espírito Santo state police, this work used a portable spectrometer in the NIR region associated with chemometric tools such as principal component analysis (PCA) and partial least squares regression (PLS) to classify and quantify possible adulterants present such as soybean oil. In addition, physicochemical tests (acidity, peroxide and ultraviolet absorption) were carried out on commercial samples. The PLS regression model built to quantify soybean oil showed a coefficient of determination (R2) greater than 0.99, showing itself to be satisfactory. The results showed that all samples analyzed had a high content of adulterant and cannot be classified as extra virgin olive oil.

Myracrodruon urundeuva (“aroeira-do-sertão”) is a species threatened with extinction due to anthropogenic exploitation. Phytochemical analysis of bark, branch and leaf extracts revealed the presence of several compounds such as flavonoids, phenols, tannins, quercetin derivatives and anacardic acids. Dereplication methodology was performed to tentatively identify 50 compounds analyzed by ultra-performance liquid chromatography coupled with an electrospray ionization quadrupole time-of-flight mass spectrometry operating in MSE mode (UPLC-QTOF-MSE). The extracts exhibited anti-tumor effect in cancer cells HCT-116 (colorectal), SF-295 (glioblastoma), HL-60 (leukemia), and RAJI (leukemia). Also, these results correlate with the principal component analysis (PCA) data that identified three distinct groups indicating, efficiently, metabolic differences between organs of M. urundeuva. Through discriminatory analysis of the orthogonal partial least squares (OPLS-DA), the variable of importance in the projection (VIP) and S-Plot, we were able to determine 30 potential biomarkers. The fingerprint of hydroethanolic extracts was correlated with the cytotoxicity assay and demonstrated a significant difference in the composition of plant extract.

Our study aimed at the development of a method based on ultra-performance liquid chromatography coupled to mass spectrometry (UPLC-ESI-MS) using selected ion monitoring for the simultaneous quantitation of the alkaloids galantamine, pseudolycorine, sanguinine and narciclasine in Amaryllidaceae species (Hippeastrum elegans, Habranthus cf. irwinianus, Hymenocallis littoralis and Griffinia nocturna). The alkaloids were extracted from dried and ground bulbs (100 mg) using liquid-liquid microextraction followed by solid phase extraction in cation exchange cartridges. The quantification method showed good linearity (correlation coefficient, R ≥ 0.9968) and selectivity for a run time of 10 min. All values were within the acceptable limits for recovery (87.5-96.2%), interday (coefficient of variation, CV%, 1.3-8.4%) and intraday precision (CV% 5.7-8.1%), except narciclasine (70%). The limits of detection and quantification were 5-100 and 20-350 ng mL-1, respectively. Our method demonstrated to be rapid, sensitive and reliable, therefore appropriate for being employed in the prospection of new source of galantamine and other alkaloids.

OBJECTIVE: To assess the incidence of intra-operative immediate hypersensitivity reactions and anaphylaxis. METHODS: A cross-sectional observational study was conducted at the Department of Anesthesiology, University of São Paulo School of Medicine, Hospital das Clínicas, São Paulo, Brazil, from January to December 2010. We developed a specific questionnaire to be completed by anesthesiologists. This tool included questions about hypersensitivity reactions during anesthesia and provided treatments. We included patients with clinical signs compatible with immediate hypersensitivity reactions. Hhypersensitivity reactions were categorized according to severity (grades I-V). American Society of Anesthesiologists physical status classification (ASA 1-6) was analyzed and associated with the severity of hypersensitivity reactions. RESULTS: In 2010, 21,464 surgeries were performed under general anesthesia. Anesthesiologists answered questionnaires on 5,414 procedures (25.2%). Sixty cases of intra-operative hypersensitivity reactions were reported. The majority patients (45, 75%) had hypersensitivity reactions grade I reactions (incidence of 27.9:10,000). Fifteen patients (25%) had grade II, III or IV reactions (intra-operative anaphylaxis) (incidence of 7:10,000). No patients had grade V reactions. Thirty patients (50%) were classified as ASA 1. The frequency of cardiovascular shock was higher in patients classified as ASA 3 than in patients classified as ASA 1 or ASA 2. Epinephrine was administered in 20% of patients with grade III hypersensitivity reactions and in 50% of patients with grade II hypersensitivity reactions. CONCLUSIONS: The majority of patients had hypersensitivity reactions grade I reactions; however, the incidence of intra-operative anaphylaxis was higher than that previously reported in the literature. Patients with ASA 3 had more severe anaphylaxis; however, the use of epinephrine was not prescribed in all of these cases. Allergists and anesthesiologists should implement preventive measures to reduce the occurrence of anaphylaxis.

Four new alkaloids, Brazoides A-D, together with three known compounds squalene, β-sitosterol and lupeol, were isolated from leaves of Justicia gendarussa. These structures were established by spectrometric techniques, mainly high-resolution electrospray ionization mass spectrometry (HRESIMS) and 1D and 2D nuclear magnetic resonance (NMR), including comparative analysis with literature values. Structural determination of the compounds, Brazoides A-D, was strengthened by molecular modeling and density functional theory (DFT) calculations to predict the NMR data and compare with the experimental NMR values of these natural products. The new compounds were tested against three human cancer cell lines (glioblastoma, prostate and colon), but none exhibited activity.

Four new epimeric spirostanol and furostanol-type steroidal saponins, (25R,S)-5α-spirostan-2α,3β-diol 3-O-β-D-glucopyranosyl-(1→4)-β-D-galactopyranosyl-(1→4)-β-D-galactopyranoside, (25R,S)-5α-spirostan-2α,3β-diol 3-O-β-D-glucopyranosyl-(1→2)-α-L-rhamnopyranosyl-(1→4)-β-D-galactopyranoside, 26-O-β-D-glucopyranosyl-(25R,S)-5α-furost-20-ene,2α,3β-diol 3-O-β-D-galactopyranoside and 26-O-β-D-glucopyranosyl-(25R,S)-5α-furost-20-ene,2α,3β-diol 3-O-β-D-galactopyranosyl-(1→4)-β-D-galactopyranoside, in addition to the known (25R,S)-5α-spirostan-2α,3β-diol 3-O-β-D-galactopyranoside, were isolated from Cestrum laevigatum. Compounds were submitted to cytotoxic activity assays using colorectal adenocarcinoma (HCT-116), human promyelocytic leukemia (HL-60), ovarian carcinoma (OVCAR-8), glioma (SF-295) human cancer cell lines, and the antimicrobial activity was evaluated against Candida parapsilosis, C. albicans, C. krusei, Pseudomonas aeruginosa, Staphylococcus aureus and Bacillus subtilis.

Abstract In some operational circumstances a fast evaluation of landfill leachate anaerobic treatability is necessary, and neither Biochemical Methane Potential nor BOD/COD ratio are fast enough. Looking for a fast indicator, this work evaluated the anaerobic treatability of landfill leachate from São Carlos-SP (Brazil) in a pilot scale Anaerobic Sequence Batch Biofilm Reactor (AnSBBR). The experiment was conducted at ambient temperature in the landfill area. After the acclimation, at a second stage of operation, the AnSBBR presented efficiency above 70%, in terms of COD removal, utilizing landfill leachate without water dilution, with an inlet COD of about 11,000 mg.L-1, a TVA/COD ratio of approximately 0.6 and reaction time equal to 7 days. To evaluate the landfill leachate biodegradability variation over time, temporal profiles of concentration were performed in the AnSBBR. The landfill leachate anaerobic biodegradability was verified to have a direct and strong relationship to the TVA/COD ratio. For a TVA/CODTotal ratio lower than 0.20, the biodegradability was considered low, for ratios between 0.20 and 0.40 it was considered medium, and above 0.40 it was considered high.

Resumo Um levantamento atualizado das plantas com sementes e análises relevantes acerca desta biodiversidade são apresentados. Este trabalho se iniciou em 2010 com a publicação do Catálogo de Plantas e Fungos e, desde então vem sendo atualizado por mais de 430 especialistas trabalhando online. O Brasil abriga atualmente 32.086 espécies nativas de Angiospermas e 23 espécies nativas de Gimnospermas e estes novos dados mostram um aumento de 3% da riqueza em relação a 2010. A Amazônia é o Domínio Fitogeográfico com o maior número de espécies de Gimnospermas, enquanto que a Floresta Atlântica possui a maior riqueza de Angiospermas. Houve um crescimento considerável no número de espécies e nas taxas de endemismo para a maioria dos Domínios (Caatinga, Cerrado, Floresta Atlântica, Pampa e Pantanal), com exceção da Amazônia que apresentou uma diminuição de 2,5% de endemicidade. Entretanto, a maior parte das plantas com sementes que ocorrem no Brasil (57,4%) é endêmica deste território. A proporção de formas de vida varia de acordo com os diferentes Domínios: árvores são mais expressivas na Amazônia e Floresta Atlântica do que nos outros biomas, ervas são dominantes no Pampa e as lianas apresentam riqueza expressiva na Amazônia, Floresta Atlântica e Pantanal. Este trabalho não só quantifica a biodiversidade brasileira, mas também indica as lacunas de conhecimento e o desafio a ser enfrentado para a conservação desta flora.
Abstract An updated inventory of Brazilian seed plants is presented and offers important insights into the country's biodiversity. This work started in 2010, with the publication of the Plants and Fungi Catalogue, and has been updated since by more than 430 specialists working online. Brazil is home to 32,086 native Angiosperms and 23 native Gymnosperms, showing an increase of 3% in its species richness in relation to 2010. The Amazon Rainforest is the richest Brazilian biome for Gymnosperms, while the Atlantic Rainforest is the richest one for Angiosperms. There was a considerable increment in the number of species and endemism rates for biomes, except for the Amazon that showed a decrease of 2.5% of recorded endemics. However, well over half of Brazillian seed plant species (57.4%) is endemic to this territory. The proportion of life-forms varies among different biomes: trees are more expressive in the Amazon and Atlantic Rainforest biomes while herbs predominate in the Pampa, and lianas are more expressive in the Amazon, Atlantic Rainforest, and Pantanal. This compilation serves not only to quantify Brazilian biodiversity, but also to highlight areas where there information is lacking and to provide a framework for the challenge faced in conserving Brazil's unique and diverse flora.

As mudanças no uso e na ocupação do solo, provocadas pelas ações antrópicas, têm gerado grandes impactos nas paisagens. Esses impactos podem ser mitigados através do monitoramento do uso e da cobertura do solo utilizando-se informações espaço-temporais das modificações ocorridas na paisagem. Este trabalho apresenta uma análise espaço-temporal detalhada da dinâmica do uso e ocupação do solo da bacia hidrográfica do riacho São Paulo, localizada na região semiárida do Estado de Pernambuco, entre os anos de 1991 e 2010. Partindo da classificação de três imagens de satélite dos anos de 1991, 2000 e 2010, foram utilizadas matrizes de transição, associadas à álgebra de mapas e métricas da paisagem. Os resultados mostraram que durante o período estudado as classes de uso e ocupação do solo passaram por uma grande transição, com a substituição progressiva das áreas de vegetação de caatinga aberta por áreas com pastagem/agricultura. Junto com o rápido crescimento da classe pastagem/agricultura, a paisagem predominante de vegetação nativa foi sendo substituída por uma paisagem cada vez mais devastada, heterogênea e fragmentada, como mostraram os índices das métricas da paisagem.
The changes in the use and occupation of land, caused by human actions, have created major impacts on the landscapes. These impacts can be mitigated by monitoring the use and land cover, using spatial and temporal information of the changes occurring in the landscape. This paper presents a detailed analysis of spatial and temporal dynamics of land use and occupation of the stream São Paulo watershed, located in the semiarid region of Pernambuco State, during the period from 1991 to 2010. Based on the classification of three satellite images for the years 1991, 2000 and 2010, transition matrices were used associated with the algebra of maps, and landscape metrics. The results showed that during the studied period, the classes of land use and occupation went through a major transition, with the gradual replacement of sparse area of 'caatinga' vegetation by grazing areas/agriculture. Along with the rapid growth of the grazing areas/agriculture class, the predominant landscape of native vegetation has been replaced by a landscape increasingly devastated, fragmented and heterogeneous, as shown by the index of landscape metrics.

Study of mechanical properties and durability of magnesium phosphate cement matrix containing grinding dust

The acid-base cements are materials developed by means of reactions between an acid component and another basic one. This type cement belongs to the family of the so called "chemically bonded phosphate ceramics" (CBPCs), new types of inorganic materials that have special physico-chemical properties and that, in contrast to most advanced ceramics, are made by processing the material at ambient temperature. In the present work, cylindrical specimens were molded (30 mm × 50 mm) with magnesium phosphate cement and grinding dust (waste from an automobile industry) which was added to ceramic matrices in varied contents (0% to 40% in mass). The influence of this addition with regard to phase formation and to mechanic resistance (axial compression and traction by diametrical compression) of the compositions (3, 7 and 28 days after the molding) was evaluated using a universal test machine. The samples of magnesium phosphate cement mortars containing grinding dust were then analyzed after being subjected to an accelerated aging test for a period of 1200 hours (50 days), equivalent to one year of natural aging. The results obtained showed a decrease in the compressive strength of the mortar between 28 days and 1 year of age but this loss in performance is not associated with the presence of grinding dust and the CBPCs proved highly satisfactory for the encapsulation of hazardous wastes.

BACKGROUND:Interferential electrical stimulation (IES), which may be linked to greater penetration of deep tissue, may restore blood flow by sympathetic nervous modulation; however, studies have found no association between the frequency and duration of the application and blood flow. We hypothesized that 30 min of IES applied to the ganglion stellate region might improve blood flow redistribution.OBJECTIVES: The purpose of this study was to determine the effect of IES on metaboreflex activation in healthy individuals.METHOD: Interferential electrical stimulation or a placebo stimulus (same protocol without electrical output) was applied to the stellate ganglion region in eleven healthy subjects (age 25±1.3 years) prior to exercise. Mean blood pressure (MBP), heart rate (HR), calf blood flow (CBF) and calf vascular resistance (CVR) were measured throughout exercise protocols (submaximal static handgrip exercise) and with recovery periods with or without postexercise circulatory occlusion (PECO+ and PECO -, respectively). Muscle metaboreflex control of calf vascular resistance was estimated by subtracting the area under the curve when circulation was occluded from the area under the curve from the AUC without circulatory occlusion.RESULTS:At peak exercise, increases in mean blood pressure were attenuated by IES (p<0.05), and the effect persisted under both the PECO+ and PECO- treatments. IES promoted higher CBF and lower CVR during exercise and recovery. Likewise, IES induced a reduction in the estimated muscle metaboreflex control (placebo, 21±5 units vs.IES, 6±3, p<0.01).CONCLUSION: Acute application of IES prior to exercise attenuates the increase in blood pressure and vasoconstriction during exercise and metaboreflex activation in healthy subjects.
