Phlebotominae (Diptera: Psychodomorpha: Psychodidae) deposited in the Museum of Zoology of the University of São Paulo

Abstract

The subfamily Phlebotominae is represented by insects commonly known as sand flies widely distributed in tropical and subtropical regions, with the Neotropical region having the greatest diversity. Only New Zealand or the Pacific Islands, excepting New Caledonia, do not present records. One means of access to the more than 1,060 described species is through the specimens deposited in biological collections, and the availability of associated data on online platforms for consultation on the part of students and researchers. This being the case, this study aimed catalogue the sand flies deposited in the Invertebrate Collection of the MZUSP (University of São Paulo Museum of Zoology). Twenty-nine species designated as type material were found being of nine holotypes, one “allotype” and 90 paratypes described from five countries in the Americas: Panama, Honduras, Jamaica, Mexico, and Brazil. In addition to these, 1,346 non-type specimens, from 12 Brazilian states, are deposited in the collection and have been included here in list form. The MZUSP sand fly collection currently houses specimens from 152 species, which represents 28% (546) of the Phlebotominae fauna of the Americas.

INTRODUCTION

Sand flies are dipterans belonging to the family Psychodidae and subfamily Phlebotominae, responsible for transmitting several pathogens, including parasites of the genus Leishmania (Lainson & Rangel, 2005). There are 1,060 known species of Phlebotominae worldwide, with 53% (546 specimens) already recorded in the Americas, of which 297 (54%) occur in Brazil (Shimabukuro et al., 2017; Andrade et al., 2020; Galati, 2023; Galati & Rodrigues, 2023). According to Galati (2018) classification, there are 23 genera, of which Bichromomyia, Lutzomyia, Migonemyia, Nyssomyia, Pintomyia, Psychodopygus and Trichophoromyia can be important in the transmission of Leishmania to mammals (Maroli et al., 2013; Akhoundi et al., 2017). The diversity and abundance of sand flies are closely connected to abiotic factors as temperature, humidity, precipitation, the biome, availability of organic material and of food sources (Quaresma et al., 2012; Oliveira et al., 2013; Parham et al., 2015; Peterson et al., 2017). In Brazil, due to the environmental conditions that favor the development of these insects, their presence is observed in forest, rural and urban environments (Almeida et al., 2010; Silva et al., 2011).

Sand flies can be found in institutional collections (Santos et al., 2014; Adams & Shimabukuro, 2018, Andrade-Filho et al., 2022), including the named type-series, composed of the type specimen which is the example from which the taxon was described (ICZN, 1999). Brazil boasts seven scientific collections that house type specimens of the Phlebotominae: (I) ”Coleção de Flebotomíneos do Instituto Evandro Chagas” (ColFleb IEC); (II) ”Coleção de Flebotomíneos” (FIOCRUZ/COLFLEB); (III) ”Coleção de Entomologia Médica da Faculdade de Saúde Pública da Universidade de São Paulo” (FSP USP/SP); (IV) ”Coleção de Invertebrados do Museu de Zoologia da Universidade de São Paulo” (MZUSP/SP); (V) ”Coleção de Invertebrados do Instituto Nacional de Pesquisas da Amazônia” (INPA/AM); (VI) ”Coleção Entomológica do Instituto Oswaldo Cruz” (CEIOC/RJ) and (VII) ”Coleção Entomológica dos Instituto Butantan” (IBut/SP). In addition to these, it is possible to obtain information on types of Phlebotominae in collections abroad (Adams & Shimabukuro, 2018).

Among the above-mentioned collections, MZUSP houses the most important collection of Neotropical Diptera, including types, and is thereby characterized as the major collection of this group in Latin America. It currently comprises around 550,000 dried mounted samples and around 500,000 samples of entomological material preserved in wet method (MZUSP, 2021). Faced with the importance of the collections for the preservation of biodiversity, and of sand flies, regarding health, we catalogued the material type and listed the non-type specimens of Phlebotominae (Diptera: Psychodidae) deposited in the MZUSP collection. METHODS

Previous information contained on the permanent slides deposited in the MZUSP collection was annotated and photographed, followed by a verification of the original descriptions found in the literature. The catalogue contains the information on these descriptions (specimen name, locality, collection method, date, and collector/legit) keeping all of the original information in the labels. The condition of the specimens was observed. The nomenclature of the types in this catalogue and the geographical distribution of the species are following Galati (2018). This catalogue was devised and organized according to previous research on other groups of insects, including sand flies (Santos et al., 2014; Adams & Shimabukuro, 2018; Falaschi et al., 2020; González et al., 2021). The other species of Phlebotominae (non-type) deposited in the collection are listed in the Supplementary Material.

The maps were devised in the QGIS program, version 3.20.2 (QGIS, Development Team, Open-Source Geospatial Foundation OSGeo). The maps represent the record for each specimen using the geographical coordinates of the localities mentioned in the labels on the deposited slides.

The types listed are arranged in alphabetical order of genus and divided according to subtribe, using the following format:

RESULTS

The MZUSP collection houses 1,446 specimens of sand flies, included in 152 species, distributed in the subtribes Brumptomyiina (16 species), Lutzomyiina (62 species), Sergentomyiina (14 species), Psychodopygina (59 species) and undetermined subtribe (1 species). The origin of the type species includes five Latin American countries and eight Brazilian states (Fig. 1). Non-type specimens from the collection include eight Latin American countries and 12 Brazilian states (Fig. 2). This catalogue presents information on types of 29 species of Phlebotominae, deposited in the Diptera collection at MZUSP. Data on type-species was incorporated into the Taxonomic Catalog of Brazilian Fauna (http://fauna.jbrj.gov.br).

However, this species was invalidated by Andrade et al. (2013), since this specimen is composed of parts of different species.

DISCUSSION

The collection of invertebrates at MZUSP is recognized as one of the greatest scientific collections in Brazil by presenting a wide diversity of species of the most diverse taxa and because of its historical character (Vivo et al., 2014). The Phlebotominae collection corresponds to 0.2% of the deposited insects, but it has 28% of the known sand fly species of the Americas. All type specimens are mounted in Canadian Balm. They are well preserved and body parts are visible, enabling specific identification. Yet, it should be noted that the number of type-species could be greater, since, after verifying data from the slides of the species Martinsmyia alphabetica, Evandromyia wilsoni, Brumptomyia cardosoi, Br. mangabeirai we observed that the information corroborates with that contained in the literature describing the species. But without the designation of the authors on the slides, it is difficult to verify their status as part of the type-series. In the same situation, we found in the MZUSP collection specimens of Br. travassossi and Pintomyia damascenoi collected by Mangabeira-Filho (1941, 1942). We believe that these sand flies could be part of the collection of more than 30 specimens described by the author. The Mangabeira’s collection was in the Entomological Collection of the Instituto Oswaldo Cruz (FIOCRUZ/CEIOC) but, until moment, it is lost (Andrade. AJ, personal communication). On the other hand, the MZSUP received some of the slides originally belonging to the USP Department of Medicine/Parasitology Standard Collection, an important collection set up and organized by Dr. Mauro Pereira Barretto, who for almost 60 years devoted himself to the study of sand flies (Shimabukuro & Galati, 2011, Andrade et al., 2013).

The species, listed in this catalogue present a wide distribution in the Americas. In Brazil, there are records of Phlebotominae in every state (Galati, 2018) and the occurrence of these insects in different regions can give indications of environmental quality, as well as information necessary for the study of the epidemiological aspects of leishmaniases (Pereira-Júnior et al., 2019). The preservation of samples in collections could be essential in studies on the dynamics of a disease in a certain region. It is possible to utilize information on labels or in genetic material banks to evaluate the seasonal dynamics of vector species, to estimate the space-time distribution of the species and also analyze competence in the transmission of pathogens (Maggi & Krämer, 2019; Thompson et al., 2021).

Entomological repositories and collections are essential for the study of insect taxonomy and systematics, above all those of medical interest, since it is possible to consult the material and compare it with the original description to check the taxonomic status of the species, for example (Andrade et al., 2013, 2014). A consultation of type material already collected and catalogued becomes an interesting alternative for the study of vectors, since there is information on taxonomic aspects, taxonomic status of the specimen and the biogeography of a specimen or group, principally of those groups that are difficult to capture (Cranston, 2005). For example, the specimen Phlebotomus breviductus Barretto, 1950, which is in the MZUP’s collection, after evaluation of the type material was ultimately invalidated by Andrade et al. (2013), thus showing the importance of constant revision of the material stored so as to discover occasional irregularities and correct them.

In this work was observed the presence of species and/or potential species that can transmit Leishmania, for example Lutzomyia longipalpis, which is the main species responsible for the transmission of Leishmania (Leishmania) infantum (Lainson & Rangel, 2005). Nyssomyia trapidoi and Ny. ylephiletor are potential vectors of Leishmania (Viannia) panamensis (Maroli et al., 2013; Dutari & Loaiza, 2014). This demonstrates the importance of preserving this type of material to support taxonomic identifications of vector species and contribute to a better understanding of the eco-epidemiological aspects in the transmission chain of leishmaniasis (Thompson et al., 2021).

In addition to the role of preserving biodiversity, the collections can provide rapid access to samples which are important and once digitalized can facilitate the propagation of information (Balke et al., 2013; Beaman & Cellinese, 2012; Hedrick et al., 2020), and promote the use of new analytical approaches to old samples to generate new knowledge, enabling more complex studies to be performed. One of the tools that can be used in studies of insects is the use of the DNA barcode to identify taxonomic specimens or groups. It is also possible to apply this type of approach in studies that involve the areas of ecology and conservation (Kress et al., 2015). The collections could thus be useful for conducting studies of taxonomic review of a specimen for example, since there is the possibility to compare deposited samples by studying vouchers and reference materials and re-use them with new research perspectives. (Camargo et al., 2015; SciColl, 2016).

For these reasons, scientific collections are indispensable in the preservation of natural history. The information contained can serve as a resource for the most diverse types of academic research, in addition to the possibility of being used for educational ends.
