Resumo A melhoria da germinação de plantas é fundamental para garantia de mudas de melhor qualidade. Assim, objetivou-se avaliar se o condicionamento de sementes com qualidade de luz (light quality - LIQ) e extrato aquoso de tubérculos de Cyperus rotundus (AEC) modula a germinação e o crescimento inicial de plântulas de Moringa oleifera. Utilizou-se delineamento inteiramente casualizado, em esquema fatorial 4x4, sendo quatro condições de LIQ (luz branca, azul, vermelha e vermelho distante) e quatro concentrações de AEC (0, 25, 50 e 100%). O condicionamento de sementes com luz vermelha reduziu o tempo médio de emergência, enquanto que as luzes azul, vermelha e vermelho extremo associadas a 50% de extrato aquoso de C. rotundus aumentaram o comprimento inicial da parte aérea e o acúmulo de pigmentos fotossintéticos. Condicionamento de sementes com luz azul induziu a formação de plântulas com menor comprimento final da parte aérea, no entanto, a aplicação de 100% de extrato aquoso de C. rotundus reverteu o menor crescimento. A luz branca associada às concentrações de 50 e 100% de fitormônios promoveu maior taxa de crescimento relativo da parte aérea de plântulas. Nossa pesquisa mostra que o condicionamento de sementes com radiação espectral de luz e extrato aquoso de tubérculos de Cyperus rotundus modula a germinação e o crescimento inicial de plântulas de Moringa oleifera. Mais trabalhos precisam ser feitos para determinar os compostos do AEC responsáveis que que atuam como fitormônios e são responsáveis pelo crescimento inicial.

Abstract Improving plant germination is essential to guarantee better quality seedlings. Thus, this research aimed to evaluate whether the seed priming with light quality (LIQ) and the aqueous extract of Cyperus rotundus (AEC) tuber could modulate the germination and initial growth of Moringa oleifera L. seedlings. The experimental design was a completely randomized in the 4x4 factorial scheme, composed of four LIQ conditions (white, blue, red, and distant red light) and four AEC concentrations (0, 25, 50 and 100%). Seed priming with red light reduced the average emergence time, while blue, red, and extreme red lights associated with 50% of aqueous extract of C. rotundus increased shoot initial length and photosynthetic pigment accumulation. Seed priming with blue light resulted in seedlings with a shorter final shoot length. However, application of 100% of aqueous extract of C. rotundus reversed this. The white light in combination with concentrations of 50 and 100% of AEC promoted a higher relative shoot growth rate of seedlings. The research revealed that seed priming with light quality and aqueous extracts of C. rotundus tubers modulates the germination and initial growth of M. oleifera seedlings. More work needs to be done to determine the responsible compounds in AEC that is responsible for priming growth as phytohormones.

Resumo A FAO estima que 30% da produção agrícola anual se perca ou se desperdice ao longo da cadeia de distribuição. O sector da restauração é responsável por 9 a 20% deste desperdício. As cantinas Universitárias servem milhares de refeições diariamente e originam desperdício devido à má gestão, ambiente dos refeitórios e comportamento dos utentes. Este estudo piloto visa examinar as principais causas do desperdício e compreender quais as atitudes dos utentes que frequentam o Restaurante universitário da ESALQ USP em Piracicaba no Brasil. Usando uma metodologia mista os resultados sugerem que existe uma grande amplitude de desperdício ao longo do ano. Os questionários aplicados junto aos usuários permitem concluir que o valores dos mesmos, a qualidade dos alimentos e a quantidade servida são fatores determinantes do desperdício. Também foi possível determinar que nos semestres finais do ano academico e quando são servidos pratos à base de proteina animal o desperdício de alimentos na ESALQ é maior.

Abstract According to the FAO about 30% of the annual food production is lost or wasted along agri-food chains. Food service wastes between 9 and 20% of this volume. Universities dining halls serve thousands of meals daily and have losses and wastage due to poor management, dining hall environment and patrons’ behaviour. Using a mix methodology this preliminary study aims to understand which are the main determinants of waste as well as the patrons’ attitudes to and perception of waste. The questionnaires applied to users allow us to conclude that patron attitudes to waste, food quality and quantity are main determinants factors of waste. It was also possible to establish that the end of the year semesters and menus based on animal protein led to high levels of food waste at ESALQ dining halls.

ABSTRACT This research aims to improve the study about the stability of truss structures used in pivot irrigation, widely used due to the growing demand for food production, when submitted to wind effects. Specifically, it aims to verify the maximum wind speed supported by an irrigation center pivot structure when submitted to the load effects caused by wind gusts perpendicular to the equipment. In order to determine these forces and their effects on the irrigation equipment, computational numerical investigations were carried out using finite element bar models validated by measuring the natural frequency of the equipment in the field. A span was designed based on the existing standards in Brazil and the dynamic response of the structure was examined under the effects of wind gusts. Models of the Brazilian standard NBR 6123/88 for structures submitted to wind effects were used in this study. The studied irrigation equipment supports wind gusts of up to 115 km/h applied perpendicularly, with a duration of 3 s, before generating toppling.

Cytomegalovirus (CMV) disease is a major cause of morbidity and mortality in solid organ transplantation. Disseminated toxoplasmosis after liver transplantation is a rare but fatal event. Serologic screening of the donor and the recipient is essential to prophylactic management, early diagnosis and therapeutic strategies to minimize the consequences of these infections. The aim of the present study was to determine the seroprevalence of CMV and Toxoplasma gondii (TG) in a Brazilian liver transplant waiting list (LTWL). Serological data were collected from 44 candidates on the LTWL between May 2003 and November 2004. Serological investigation of antibodies IgM and IgG against CMV (anti-CMV) and TG (anti-T. gondii) was performed using fluorometry commercial kits. IgG anti-CMV was positive in 37 patients (94.9%) out of 39 available results. There were not IgM anti-CMV positive results. Out of 36 analyzed patients, 22 (61.1%) presented positive IgG anti-T. gondii and none had positive IgM anti-T. gondii. The high CMV seroprevalence among our LTWL reinforces the need for appropriate protocols to avoid related complications, like reactivation and superinfection by CMV. Environmental and drug prophylactic strategies against primary infection and reactivation, as well as early diagnosis and treatment of toxoplasmosis complications, are essential for the good outcome of transplant patients.

The batch copolymerization of styrene (St) and methyl methacrylate (MMA) in the presence of polybutadiene (PB) is theoretically and experimentally investigated. A mathematical model that enables to calculate the main global variables and the macromolecular structure of the evolving polymer mixture is presented. The model is an extension of that developed by Estenoz et al. (1999) for the homopolymerization of St in the presence of PB. It was validated with experiments that considered diluted solution and bulk copolymerizations carried out at a constant low temperature (65 º C), with THF as solvent, benzoyl peroxide as initiator, and a St-MMA ratio close to the azeotropic composition. For comparison, the solution copolymerization without PB was also considered. All experimental runs were adequately simulated using a single set of kinetic parameters.

Vanadium oxide supported on hydrotalcite-type precursors was studied in the oxidation of isopropanol. Hydrotalcites with different y = Mg/Al ratios were synthesized by the method of coprecipitation nitrates of Mg and Al cations with K2CO3 as precipitant. The decomposition of these hydrotalcite precursors at 450°C yielded homogeneous MgyAlOx mixed oxides that contain the Al+3 cations totally incorporated into the MgO framework. The materials were characterized by chemical analysis, BET superficial area, X-ray diffraction, temperature-programmed reduction (TPR) and the reaction of isopropanol, a probe molecule used to evaluate the acid-base properties. The results of TPR showed that the reducibility of V+5 decreased with the increase in magnesium loading in catalysts. The X-ray diffraction patterns of Al-rich hydrotalcite precursors showed the presence of crystalline phases of brucite and gibbsite. It was shown that chemical composition, texture, acid-base properties of the active sites and also Mg/Al ratio strongly affect the formation of the products in the oxidation of isopropanol. The Al-rich catalysts were much more active than the Mg-rich ones, converting isopropanol mainly to propylene.

The aim of this paper was to evaluate the immune reconstitution of HIV-1 patients subjected to highly active antiretroviral therapy (HAART) for two years or more according to CD45RA and CD45RO cell count; determination of IL-2, IFN-gamma, IL-4, IL-10 and TNF-alpha serum levels; CD4+ T and CD8+ T lymphocyte count; and plasma viral load (VL) determination. For this purpose, a cross sectional study was carried out in the Tropical Diseases Area, Botucatu School of Medicine, São Paulo State University, UNESP, Botucatu, São Paulo, Brazil. Between June 2001 and April 2002, 37 HIV-1 infected patients were evaluated, 13 with treatment indication but untreated (G1), 9 subjected to HAART for 5-7 months (G2), and 15 treated for two years or more (G3); both treated groups used medication regularly and without failure. Forty-nine normal individuals were studied as controls (GC-1 and GC-2). There was a tendency (p<0.10) for the predominance of two nucleoside reverse transcriptase inhibitors (NRTI) associated with one non-nucleoside reverse transcriptase inhibitor (NNRTI) regimen in G2; and two NRTI associated with a protease inhibitor (PI) in G3. Statistical differences between groups were seen for CD45RA (G1<[G3=GC-2]; p<0.05) and CD45RO (G1<GC-2<G3; p<0.01) cells, and CD4+ T lymphocyte count (G1<G3; G2-intermediate; p<0.05), VL determination (G1>[G2=G3]; p<0.001), TNF-alpha serum determination ([G1>G3; G2=intermediate]>GC-1; p<0.001), IL-2 (G1<[G2=G3=GC-1]; p<0.01), IFN-gamma ([G1=GC-1]<[GC-2=G3]; p<0.001), IL-4 and IL-10 ([G1=G2=G3]>GC-1; p<0.001), serum cytokine profiles, with a higher proportion of subtype 2 in G1 and mature subtype 0 in G2 and G3 (p<0.005). There was no statistical difference for CD8+ T lymphocyte counts (G1=G2=G3; p<0.50). Consistency was seen between positive correlations of profile 1 definer cytokines (IL-2 and IFN-gamma), CD45RA and CD45RO cells, and CD4+ T lymphocyte counts and between positive correlations of profile 2 definer cytokines (IL-4 and IL-10) with TNF-alpha, and VL. The negative correlations were also consistent as they expressed the inverse of the positives. The variables with the highest number of correlations were IL-2, IFN-gamma, and VL, followed by CD45RA and CD45RO cells, and IL-10. The variables with the lowest number of correlations were CD4+ T and CD8+ T lymphocytes. The results express the partial but important immune reconstitution in HIV-1 infected individuals with the interference of HAART and the importance of cytokines especially IL-2 and IFN-gamma, and CD45RA and CD45RO cells as surrogate markers of this reconstitution.

Seventy-nine HIV-1 infected patients were studied in three groups: Group G1 - 11 patients with no antiretroviral therapy; G2 - 40 patients undergoing antiretroviral therapy, 33 with only two nucleoside reverse transcriptase inhibitors (NRTI), and seven with two NRTI and one protease inhibitor (PI), all with viral load (VL) equal or higher than 80 copies of plasma RNA/ml; Group G3 - 28 patients, 23 on highly active antiretroviral therapy (HAART), 18 with two NRTI and one PI, and five with two NRTI and one non-nucleoside reverse transcriptase inhibitor (NNRTI), the remaining five with combination of two NRTI. All G3 patients had undetectable viral load for at least the past six months. The control group (Gc) included 20 normal blood donors without clinical complaints or signs of disease and negative for anti-HIV-1/2 antibodies. Serum cytokine levels pg/ml (TNF-alpha, INF-gamma, IL-2, IL-4, and IL-10) were determined in all patients including controls. CD4+ T and CD8+ T lymphocyte counts were made in the 79 patients by flow cytometry; VL determination was by NASBA technology. Analysis of results showed that the number of CD4+ T and CD8+ T lymphocytes were higher in G2 than G1, while VL was 0.5 log lower. G3 patients had similar lymphocyte values to G2, however they were chosen for G3 because their VL was undetectable, different by 4.0 log to G2. These results show the effect of antiretroviral treatment in G2 and G3 patients with better performance in the latter. Statistical difference was seen between the three groups and controls for serum cytokine behavior: TNF-alpha [H=48.323; p<0.001;(G1=G2=G3)>Gc]; INF-gamma[H=28.992; p<0.001; (G1=G2=G3)>Gc]; IL-4[H=48.323; p<0.001; (G1=G2=G3)>Gc]; IL-10[H=47.256; p<0.001; (G1=G2=G3)>Gc. There was no statistical difference in IL-2 values between all groups (H=6.071; p>0.10; G1=G2=G3=Gc). In absolute values however, G3 showed slightly lower TNF-alpha, IL-4, and IL-10, and higher INF-gamma and IL-2, to G1 and G2. This suggests a better performance in G3 patients, especially in IL-2 behavior. For cytokine profile, the three groups showed mature Th0 subset. In G1 72.73% were mature Th0, and 27.27% Th2; G2, 72.50% mature Th0, and 27.50% Th2; and G3, 89.29% mature Th0, and 10.71% Th2. There was no statistical difference between groups (chi22=3.014; p>0.10; G1=G2=G3). Statistical difference was seen between G2 and G3 for antiretroviral regimes used (chi21=27.932; p<0.001; G3>G2); HAART with PI predominated in G3, suggesting that it was responsible for this better performance. Linear correlation between pairs of variables was made with patient groups only, excluding controls. This was made separately for G1 and G2, 51 patients with detectable VL, and G1, G2, and G3 also including those with undetectable VL. The results showed a strong positive correlation between TNF-alpha and IL-4; TNF-alpha and IL-10; INF-gamma and IL-2; IL-4 and IL-10; IL-2 and CD4+. Weak negative correlation was seen between IL-2 and VL. Considering all correlations together, we found that IL-2 had the most correlations - eleven - strong, weak, positive, and negative; it was the only one that correlated with CD4+ (positively) and LV (negatively). The number of correlations allowed us to evaluate qualitative aspects such as IL-2 correlated positively with INF-gamma and CD4+ and negatively with LV; this somehow expresses the compatible profile with subset Th1, which could signify a tendency towards immune response recovery. Determination of cytokine serum values, especially IL-2, could be useful in follow-up of HIV-1 infected patients under HAART together with CD4+ and VL count.
